Abstract
UV (Ultra Violet) spectrophotometric and HPLC (High Performance Liquid Chromatography) methods have been developed and validated for analysis of diclofenac sodium in plasma male wistar rat in vitro. The developed methods were validated with respect to linearity, accuracy, and precision. The UV spectrum recorded using aquabidest as solvent and the wavelength 276 nm was selected for the determination of diclofenac sodium. RPHPLC analysis was carried out using shim pack CLC ODS column and mobile phase composed of methanol and acetate buffer (70:30 v/v), pH adjusted to 4.2 at a flow rate of 0.7 mL/min. The developed methods illustrated excellent linearity (r 2 > 0.99) in the concentration range of 5-27.5 μg/mL and 1-7 μg/mL for UV spectrophotometric and HPLC methods, respectively. Precision (%RSD < 1.0) for all methods, and mean recoveries were found 109.4% for UV spectrophotometric method and 105.5% for HPLC method which shows accuracy of the methods. Determination of diclofenac sodium generic and brand-name tablets in male wistar rat plasma in vitro was in the range of 90-110% for UV spectrophotometric method as well as HPLC method.
