Abstract
This research aimed to: (1) explore and identify antagonist bacterial of Bacillus sp. from maize rizospher, (2) identify of pathogenic bacteria stewart wilt in maize crop, (3) test the effectivennes antagonist bacterial of Bacillus sp. against pathogenic bacterial by in vitro; (4) know the mechanism of inhibition antagonist bacterial of Bacillus sp. against stewart’s wilt. The research was carried out by in vitro at the Laboratory of Plant Protection, Faculty of Agriculture, Jenderal Soedirman University. Exploration of antagonistic bacteria and Stewart wilter pathogens was taken at the Grendeng maize crop. The research was conducted from October to December 2017. The research completely randomized design was used with 9 replicates and 3 treatments. The bacterial isolates obtained were identified by key identification and then Koch postulates were tested. The variables observed were morphological test, biochemical test such as gram test, catalase test, oxidase test, motility test, starch hydrolysis test, anaerobic test, growth physiology test at 40˚C, pathogenicity test and in vitro test. Results of the research showed that Bacillus sp. found in the corn rhizosphere is characterized by circular (bulbous) and punctiform (rounded) bacterial colonies, variations in margins of the entire (fine) and lobate (wavy or jagged), dull white, not slimy, gram-positive bacteria, staggered and motionally immobilized, identification of Stewart wilt pathogens having yellow colonies on NA medium, bright yellow in Kings'B medium, having slimy, non-motile, sparkling, gram negative, facultative anaerobic, -motil, positive catalase, negative oxidase, positive starch hydrosis, 40˚C negative temperature growth test, and positive pathogenicity test, Bacillus sp. the most effective in inhibiting Stewart's wilter pathogen bacteria was Bacillus Bn1 with an area of 3.29 mm inhibition zone, the barrier mechanism of Bacillus sp. against Stewart wilt pathogen bacteria is bacteriostatic.
