Abstract
Ten isolates of endophytes bacteria have been tested as biofertilizer due to their ability to synthesize growth hormone IAA and fixate nitrogen. They are intended to be produced on industrial scale using fermentation principle. This research aim to discover pH and temperature combination that yield the best fermentation result. Three of ten isolates (IM1, IM10 and IM25) were inoculated in fermentor on M63 medium at combination of three pH levels (6.0, 6.5 and 7.0) and three temperature levels (28, 32 and 36 C) of nine total combinations using randomly complete factorial design. Fermentation is carried out for 18 hours at each combination. Samples are taken every two hours which absorbances are measured using spectrophotometer at λ 420 nm. Optical Densities (OD) are analyzed using quadratic regression, analysis of variance along with Tukey’s HSD test (α= 5%) and principal component analysis to discover the best combination. The result showed that combination of pH 7.0 and temperature 32 C as the best combination. OD maximum is obtained at 1.852 using quadratic regression method. As for principal component analysis, this combination produces highest score of 5.188 on behalf of two components of PC1 and PC2 with 86.97% cumulative variability.
